Basic evaluation toward radioimmuno-detection and-therapy to differentiated thyroid cancer

In an immunostaining study, a human monoclonal antithyro-globulin antibody, VB5, was shown to react the antigen on primary cultures of human thyroid cancer derived from surgical specimens.The antibody was then radiolabeled with I-125, and tested in a inding assay using the same cultured cells as in the staining works. Although measurable, mean specific binding was low at about 1% of the total radioactivity of the ladled antibody in multiple experiments. Therefore, a cocktail of antibodies against various epitopes seemed necessary to perform efficient radioimmunodetection.Despite extensive trials, we failed to establish a cancer cell line which synthesize thyroglobulin. Moreover, two cell lines provided from abroad, though reported to produce the antigen, turned out to be negative in immunostaining. We may need some molecular biological techniques to obtain a transplantable cell line, which will lead to an animal model of differentiated thyroid cancer.
