The IKK/NF-κB signalling pathway regulates cellular responses to infection, inflammation and stress. In the epidermis, IKK subunits display differential functions. IKK1 is required for keratinocyte differentiation and formation of a stratified epidermis. In contrast, IKK2 and NEMO are not needed for epidermal development, but play a crucial role in the regulation of the immune homeostasis in the skin. In the context of this project proposal, we aim to investigate the cellular and molecular mechanisms by which IKK1, IKK2 and NEMO mediate their differential functions in epidermal keratinocytes. For these studies we are using conditional targeting of IKK1, IKK2 and NEMO in mice.
