(1) Cerebral endothelial cell damage induced by sodium arachidonate in vitroCytotoxic effect of sodium arachidonate on cultured microvascular endothelial cells derived from mongolian gerbil brain was measured by specific release of ^<51>Cr. Protective effect of cyclooxygenase and lipoxygenase inhibitors (indomethacin and nordihydroguaiaretic acid respectively) on cultured endothelial cells was also examined.Sodium arachidonate injured endothelial cells at the minimal concentration of 3mug/ml, and the cytotoxicity was dependent on the concentration of sodium arachidonate and incubation time. Nordihydroguaiaretic acid inhibited the endothelial injury by sodium arachidonate. On the other hand indomethacin had no protective effect on the endothelial cells. It is posturated that the endothelial cell injury was caused by lipoxygenase product.(2) Effect of arachidonate derivatives on cerebral endothelial permeability in vitroEndothelial cells derived from songolian gerbil brain were cultured on collagen coated membrane filter, and the effect of arachidonate derivatives on passage of FITC-albumin through endothelial cells was examined.Prostaglangin E_2 and F_2 alpha did not increase the permeability of endothelial cells. 15-HPETE increased the endothelial permeability slightly (p< 0.05). Leucotriene C_4 markedly increased the endothelial permeability (p<0.001). Leucotrienes may play an important role in the development of ischemic brain injury by increasing the cerebral microvascular endothelial permeability.
(1)花生四烯酸钠对体外培养的长爪沙鼠脑微血管内皮细胞的细胞毒性作用采用特异性释放~(13)Cr的方法,观察花生四烯酸钠对长爪沙鼠脑微血管内皮细胞的细胞毒性作用<51>。环氧合酶和脂氧合酶抑制剂吲哚美辛和去甲二氢愈创木酸对培养的内皮细胞的保护作用也进行了研究,花生四烯酸钠在3 μ g/ml的最低浓度下对内皮细胞有损伤作用,其细胞毒性与花生四烯酸钠的浓度和孵育时间有关。去甲二氢愈创木酸可抑制花生四烯酸钠对内皮细胞的损伤。吲哚美辛对内皮细胞无保护作用。推测内皮细胞损伤是由脂氧合酶产物引起的。(2)花生四烯酸衍生物对脑内皮细胞通透性的影响将沙土鼠脑内皮细胞培养在胶原膜滤器上,观察花生四烯酸衍生物对FITC-白蛋白通过内皮细胞的影响,前列腺素E_2和F_2 α对内皮细胞通透性无明显影响。15-HPETE可轻度增加内皮细胞通透性(p< 0.05)。白三烯C_4能显著增加内皮细胞通透性(p<0.001)。白三烯可能通过增加脑微血管内皮细胞通透性在缺血性脑损伤的发生发展中起重要作用。
