Using transfectant cells by GM2/GD2 synthase cDNA,we have analyzed the relationship of modified cerbohydrates with cellular proliferation, and obtained results as summarized below.1. B16 melanoma lines newly expressing GM2 showed earlier reduction of cell growth comparing with non-expressing lines, but tumor growth in vivo did not show definite difference between GM2-expressants and non-expressants.2. When we analyzed the cell adhesion to extracellular matrix, GM2-expressing cell lines showed high rate of adhesion to collagen type IV,fibrinectin and laminin, suggesting the promoting effects of GM2 in cell adhesion.3. In the adhesion experiments, addition of anti-GM2 antibody decreased the adhesion of GM2-expressing cells, indicating the direct role of GM2 in cell adhesion.4. In Northern blotting, c-myc mRNA was expressed more in the GM2-expressing cells than in non-expressants.13EA06 : Thus, newly expressed GM2 affects not only cell proliferation and adhesion, but ontracellular molecules relevant with cell proliferation.
