The SCC antigen (SCCA) is expressed in normal epithelium and the serum level of SCC antigen is elevated not only in cancer patients but also in benign inflammatory diseases such as bronchial asthma and pneumonia. However, the molecular mechanism of the overexpression of SCCA is still unclear. We have previously reported that T lymphocytes surrounding head and neck sqouamous cell carcinoma (HNSCC) tissue are overexpressing SCCA and are responsible for the serum elevation of SCC antigen. TNF-alfa stimulation of T-lymphocytes induced SCC antigen expression suggesting that cytokines expressed by the tumor cells induce SCCA expression. Continuous research is on going regarding the mechanism of SCCA expression. We have also transfected SCCA (SCCA1) to cultured HNSCC cells and showed that SCCA1 expression significantly inhibited the in vitro invasion in matrigel assays. Furthermore, the in vivo growth and invasion in nude mice was also inhibited by SCCA1 expression.
