项目摘要

日本における乳癌、前立腺癌の発生率は欧米の1/5〜1/10と低い。しかし、日本人の食事と体格の欧米化と共に増加をつづけている。若い時から性腺のない人における乳癌、前立腺癌の発生率は、正常人の1%以下であるから、性ホルモン依存性癌の発生には性ホルモンが最大の影響を与える。また、発生促進は、潜伏癌から臨床癌への進展が鍵をにぎると考えられている。したがって、発癌における潜伏癌の進展の意義、性ホルモンの演じる役割を明らかにし、乳癌、前立腺癌の増加を抑制する方法を見出す。1.ヒト前立腺癌の発生率には,潜伏癌→臨床癌への進展の割合が鍵になることが明らかになっている。しかし、好い発癌実験系がないために、促進要因は不明のままである。伊東等は、F344ラットに3,2'ーdimethyl-aminobiphenylを発癌剤として投与し、生理量と元の10倍の薬理量のアンドロゲンをプロモーターに使用した。薬理量のアンドロゲンだけで40%のラットのdorsolateral prostateに浸潤癌が生じた。ventral prostate には潜伏癌が両群で同様に(30%)発生した…伊東、杉村。2.乳癌でも、潜伏癌→臨床癌への進展がその発生の鍵になると考えられている。しかし、潜伏癌の実態は不明である。坂元等は、TO非浸潤癌を潜伏癌と考え、潜伏癌は臨床癌の10ー20倍以上存在すること、エストロゲンがその進展を促進することを示した…坂元、寺田、横路。3.性ホルモンによる癌増殖促進は、増殖因子を介することが実証された。マウス乳癌をモデルに用いた…松本、佐藤。4.潜伏癌の病理診断に有用な癌遺伝子関連物質等のマーカーは見出せなかった…坂倉、広橋、西河。
