当施設で得られた子宮体癌手術材料28例をAMeX固定パラフィン包埋ブロックとし、nm23H1とnm23H2のそれぞれのモノクローナル抗体を用いて、免疫染色にて発現の局在と発現強度を、Western blottingにて発現量を定量し、転移など臨床病理学的因子との相関を検討した。nm23H1とnm23H2の免疫染色の結果(1)子宮癌細胞の細胞質に一様に発現し、子宮内膜間質および子宮筋層には発現しなかった。(2)nm23H1,nm23H2ともに、高分化癌では中分化癌に比べ有意に発現が強く(P<0.05)、低分化癌では発現の強いものと弱いものと2種類存在した。(3)転移、腫瘍径、子宮筋層浸潤、エストロゲンレセプターとプロゲステロンレセプターの発現、閉経の有無との相関はなかった。2.nm23H1とnm23H2のWestern blottingの結果(1)免疫染色の結果と同様に、nm23H1,nm23H2ともに、高分化癌では中分化癌に比べ有意に発現量が多く(P<0.05)、低分化癌では発現量の多いものと少ないものと2種類存在した。転移、腫瘍径、子宮筋層浸潤、エストロゲンレセプターとプロゲステロンレセプターの発現、閉経の有無との相関はなかった。(2)同一症例での発現を免疫染色の結果と比較したところ、評価の一致したものはそれぞれ69%、66%であり、不一致例では免疫染色の方が強く評価する傾向にあった。

When 28 cases of uterine cancer surgical materials were used, AMeX fixation, nm23H1 and nm23H2 were used to detect the presence of local antibodies, immunostaining, Western blotting, quantification, and migration of clinical pathology factors.
