At the completion of flap reflection, the soft tissues are placed in contact with the root surface. Epithelial and connective tissue reattachment is complete at 3 weeks. Freely mobile cells of the gingival lamina propria include fibroblast, mast cells, and a small number of mononuclear and polymorphonuclear leukocytes. The most numerous cell type is the fibroblast. Human gingival fibroblasts (HGFs) produce connective tissue substances including collagen, , proteoglycans, and elastin, and essentially maintain the integrity of gingival lamina propria. The present investigation was conducted evaluate the effect of human gingival fibroblasts conditioned medium (HGF-CM) to chemotaxis of human periodontal ligament fibroblasts (HPLFs), HGF and human alveolar bone cells (HABCs), and human gingival epithelial cells (HGEs).To determine whether HGF-CM was chemotactic, and/or chemokinetic for fibroblast, a checcurboard analysis of the Zigmond-Hirsch type was performed wherein different concentrations of HGF-CM were added to upper (cell) and lower (test) compartment of modified blind-well Boyden-type chamber, and migration of fibroblasts was measured. This analysis suggests that HGF-CM induced chemotactic migration of HGF and HPLF,because the cells migration increased in the presence of positive gradient, i.e.a higher of 175ng/ml concentration of the peptide in the lower compartment than in the upper compartment. HGF-CM consistently increased migration of HABC and HGE at concentration higher of 50% HGF-CM,similar result of HPLF-CM (100%) which was positive control. The data suggested that HGF-CM may contribute to the development of periodontal regeneration.
