日本人における早期発症型歯周炎患者および成人性歯周炎患者と健常者とのSNPsの比較により、多因子性疾患である歯周炎の発症および進行に関与する感受性遺伝子を検索することを目的とした。歯周炎を主訴とする早期発症型歯周炎患者12名および成人性歯周炎患者9名を対象とし、歯周炎の臨床所見がないもの19名をコントロールとした。末梢血より、ゲノムDNAを抽出し、JSNPデータベースより、IL1やTNFα等の歯周炎候補遺伝子59SNPsと候補遺伝子以外の約600SNPsについて、TaqMan PCR法によりダイビングを行い、遺伝統計学的解析を行い、歯周炎との関連について検討した。研究を行うにあたり、『ヒトゲノム・遺伝子研究に関する倫理指針』に従い、倫理的配慮のもとに研究を実施する。歯周炎候補遺伝子のSNPs遺伝統計学的解析の結果、早期発症型歯周炎患者とコントロールでは、CTSGとTRAF1で、成人性歯周炎患者とコントロールでは、IL10RBとMMP9で遺伝統計学的有意差が認められた。また、早期発症型歯周炎群と成人性歯周炎群では、IFNGにおいて有意差が認められた。また、歯周炎候補遺伝子以外についても、疾患群とコントロールとで遺伝統計学的有意差が認められた。SNPsのみの解析からは、有意差が認められたものが歯周炎の原因遺伝子であると断言はできないが、早期発症型歯周炎においては、CTSGが、成人性歯周炎においては、IL10RBとMMP9が疾患の遺伝マーカーとなる可能性が考えられる。現在さらに、サンプル数および解析するSNPsの数を大幅に増やし、SNPs解析による構造面からの歯周炎感受性遺伝子の解析を行うと同時に、歯周炎の病変歯肉から、mRNA発現解析を行っており、歯周炎との関連について機能面からの解析を行っている。
