マウス間葉系幹細胞KUSA-A1(A1)のデコリン発現をノックダウンした細胞株(siDT)を用いて、骨分化過程でのデコリンの関与と機能を解析した。siDTを骨芽細胞分化培地で誘導しても骨芽細胞へ分化せずに脂肪細胞へと分化した。A1およびsiDTをディフュージョンチャンバーに充填してマウス腹腔内に、あるいは、単独またはコラーゲンゲルとともにマウス皮下に移植して異所性骨化を生体内へ移植したところ、A1は腹腔内あるいは皮下で骨芽細胞へと分化したが、siDTは骨分化マーカーの産生が低いことや、組織学的解析結果と総合して骨分化は起きていないと判断した。以上の結果は、デコリンが間葉系幹細胞の骨分化への方向付け因子であることを強く示唆する。
