本年の実験では、BALB/cヌードマウスを用いて、脂肪幹細胞を精製し、マウスの背部に皮下注入する実験手技を確立した。当初はマウスの両側鼠径部から脂肪を採取する予定であったが、実際には鼠径部からは少量の脂肪しか採取できないことが判明したため、腹腔内の腸間膜より採取する方針とした。この時点で、ドナーマウスを生存したまま管理することが困難であり、自家脂肪由来の脂肪幹細胞の実験手技確立には更なる工夫を要することが明らかとなったが、まずは、本実験手技の確立を目指すこととし、ドナーマウスは、プロトコルに従って、安楽死させることとした。採取した脂肪を、十分に粉砕・撹拌し、0.1％コラゲナーゼを溶解したリン酸緩衝液を加えて、37℃温浴槽で処理後、ダルベッコ改変イーグル培地（DMEM）を十分量加えて、酵素を失活させ、遠心分離して培地を除去する。これにより、脂肪幹細胞を含む間質細胞集団が得られる。しかし、この操作により得られる細胞塊は極少量のため、背部に皮下注入するには不十分な量であった。注入量に十分な細胞数として、106～7個の細胞集団と設定し、上記操作で得られた細胞塊に、DMEMを加えてインキュベーター内で数日間培養を行い、間質系細胞の増殖を確認した。十分に細胞が増殖したところで、トリプシンで細胞を剥がし、この溶液を少量採取して顕微鏡下に細胞数をカウントした後、遠心分離を行い、細胞塊を単離。これに、0.1ml中に106の細胞が入る量となるよう、リン酸緩衝液を加えて、注入薬の作成が完了する。この注入薬を、レシピエントマウスの背部皮下に3～4ヶ所、1ヶ所につき0.1mlずつ注入し、同部位の観察を行った。本年度は、計10匹のレシピエントマウスに注入実験を行ったが、本年度中に、注入部位における発毛は観察されなかった。本年中は、上記実験プロトコルの確立、及び、その手技の習得を達成した。
