This project is aimed at developing a new optical recording system for the simultaneous recording of the electrical activity and the metabolism in the heart preparation. The system is composed of four main parts, including 1) optics and a recording chamber, 2) a detection system with a multiple channel recording system for the voltage-sensitive dye-recording of electrical activities, 3) the second detection system with a CCD camera for recording the metabolism of cardiac cells and 4) a computer for analyzing experimental data. The preparation chamber is placed on the stage of a microscope, in which a beam splitter is build, mounted on the vibration-isolation table. Two kinds of optical detection system are mounted to the microscope : a 16 X 16- or 12 X 12-element photodiode array for the extrinsic signals corresponding to the electrical activities, and a cooled CCD camera for the intrinsic fluorescence of NADH reflecting the metabolism of the cardiac cells. Data acquired using these detectors are transferred to a graphic workstation via LAN (local area network) to analyze the data visually. In the physiological experiments, right atrial preparations dissected from the adult rat heart are used. The preparations are stained with a merocyanine-rhodanine dye (NK2761), and a bathing medium containing 2,3-butanedione monoxime is used to reduce the contraction-related light scattering. Using this system, the relationship between the conduction pattern of the excitatory wave and the metabolic state of cardiac muscle is analyzed.
