PROJECT SUMMARY
PROTEOMICS & METABOLOMICS CORE
The goal of the Moffitt Proteomics & Metabolomics Core (P&MC) is to provide state-of-the-art equipment, cutting-
edge services, comprehensive education, and hands-on training in proteomics and metabolomics techniques.
P&MC was launched in 2003 to provide proteomic profiling. In 2016, metabolomics services were added to
support cancer metabolism, a key priority area of Moffitt’s Research Strategic Plan. Highly trained and
experienced core staff provide domain knowledge, customer service, and extensive support from project
conception to publication, leveraging cutting edge liquid chromatography- and gas chromatography-mass
spectrometry platforms complemented by diverse sample preparation techniques and data analysis strategies.
P&MC provides Member access to highly trained expert staff, leading edge proteomics and metabolomics
instrumentation, software tools, and the capability to perform appropriate experiments to address their
hypotheses. P&MC activities towards its goals are organized into three Specific Aims:
Aim 1: To provide proteomics and metabolomics expertise and resources to support Moffitt Member research.
Aim 2: To provide services, support publications and grants, and adapt to Member needs in cancer proteomics
and metabolomics, by developing novel capabilities that support the diverse research of Moffit CCSG Programs.
Aim 3: To educate and train Moffitt faculty, trainees, laboratory staff and the community on the theory and
practice of proteomics and metabolomics in cancer research.
P&MC significantly contributes to Member science at Moffitt. For example, P&MC worked with other Shared
Resources (e.g., MGC, BBSR) to integrate data supporting key proteogenomics and proteometabolomics
projects. Other key areas include: 1) development of tools to dissect the response to targeted therapy and assess
drug resistance; 2) increased capability to study cancer metabolism at the cellular and organism level, and 3)
sensitive and specific quantification of structurally similar proteins. This contribution is evidenced by P&MC
supporting 79 Members over the last five years, a 30% increase in Member usage since 2016. Members used
P&MC services for highly multiplexed measurements of proteins and metabolites to assess cancer phenotypes,
complementing traditional cancer biology approaches, leading to 50 publications (15 high impact). In FY20,
P&MC supported 32 Members in four Programs (CBE 31%, CE 6%, MM 38%, IO 25%) with 96% of those users
holding peer reviewed funding. This represented 95% of all core usage. Future priorities of P&MC are to maintain
exceptional service, improve quantitative proteomics, enhance metabolomic services, expand support of diverse
biomarker initiatives, and further build on pan-omic data analysis pipelines to enhance rigor and reproducibility,
and to maximize the knowledge gained from proteomic and metabolomic investigations.
