Neurotransmitter release from presynaptic vesicles at the synapse allows neurons tocommunicate in the brain. However the detailed spatial arrangement of differentfunctional types of synaptic vesicles within the synapse remains unknown. We will usestate-of-the-art super-resolution microscopy, including multi-colour, live and 3DSTORM/STED microscopy, and expansion microscopy, together with developing newcomputational image analysis approaches, to address this question. We will then usethese approaches to understand how this changes during synapse development, inresponse to neuronal activity and when key synaptic proteins are perturbed.
