This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. This is a non-randomized natural history non-treatment study in which a total of 800 subjects including 200 normal controls, 400 individuals with mild cognitive impairment, and 200 subjects with mild Alzheimer's Disease (AD) will be recruited at approximately fifty sites in the United States and Canada for longitudinal follow up. This research study is designed to look at the usefulness of imaging studies and biomarker tests, together with measurements of memory, thinking and daily functioning, in the future conduct of studies that will focus on the identification and treatment of Alzheimer's disease at an early stage. This study aims to produce data to be stored for these kinds of future research studies. For this project we will be recruiting at total of 20 subjects as listed below: This study will look at three different groups of volunteers: 1) Normal Control group: Volunteers with no apparent memory problems that will be studied for 36 months. Four subjects from this group will be recruited 2) Mild Cognitive Impairment (MCI) Group: volunteers with mild memory problems as determined by a series of scores on memory and cognitive tests, (referred to in this study as "mild cognitive impairment" - MCI) that will be studied for 36 months. Eight subjects will be recruited 3) Alzheimer's disease group: volunteers with mild Alzheimer's disease that will be studied for 24 months. Four subjects will be recruited, and four study partners to accompany them. All Participants must have an individual (spouse, friend, or relative), called a "study partner," who is willing to: accompany participants to all of the study visits and communicate changes in participant's health status over the period of this study. All subjects will have neuropsychological testing annually and a 1.5 Tesla brain MRI. A subset of subjects (N = 4) will receive 2 lumbar punctures (baseline and one year) and another subset of subjects will get a 3T MRI scan at a subsequent visit.
