This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Our goal is to better understand the role of obesity and exercise in cardiovascular health in children. As the atherosclerotic process can begin early in life, it is crucial to understand factors that can significantly impact this process. In adults, elevated post-prandial triglycerides (TGs) are an independent risk factor for coronary heart disease, and increased post-meal lipoprotein-derived remnant-like particles (RLP) are particularly atherogenic. Obesity in adults exacerbates post-prandial lipemia whereas exercise ameliorates this. Studies in this application will determine if this is also true in children. The post-prandial lipid response to a moderately high fat meal will be compared in obese and lean children over a 6 hr period with blood sampled every hour. Assessments will be conducted in the morning on two occasions; after a moderate exercise bout the afternoon before, and after no exercise. Primary outcomes will include post-prandial TGs, triglyceride-rich lipoprotein subfractions and associated RLP levels. Secondary outcomes will include circulating changes in cholesterol, apolipoprotein B, insulin, glucose, free-fatty acid, glycerol, and whole-body lipid oxidation. This study will provide preliminary data to more comprehensively assess factors that may impact cardiovascular health in childhood, including diabetes and ethnicity, and interventions that may improve this, such as chronic exercise alone or wi
