GENETIC VARIATION IN ACUTE DRUG EFFECTS

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The goal of this study is to determine the genetic basis of variations in acute responses to caffeine in non-chronic caffeine users. We have recently found that with a certain polymorphism of the A2A adenosine receptor gene are more likely to expreience anxiety after an acute dose of caffeine (300 mg caffeine citrate, equivalent to about 150 mg caffeine or 1 8-ounce cup of coffee). We now wish to replicate and extend these findings by testing subjects with both a lower and a higher dose of caffeine. It is hypothesized that variations in the subjective effects of caffeine will be realted to polymorphisms of the adenosine receptor, when plasma caffeine concentration is controlled.
