This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The specific aim is to test the hypothesis that the phenotype of an activating variant of the 17b-HSD5 gene depends on interactions with LH, insulin resistant hyperinsulinemia, and FSH dependent granulosa cell factors. We will begin by determining the testosterone response to the LH analog human chronic gonadotropin (hCG) in PCOS and control women of known 17b-HSD5 genotype. Then we will determine the effects of suppressing gonadotropins with a GnRH agonist, suppressing insulin with a thiazolidinedione (TZD), and stimulating granulosa cell function with FSH. The effects of suppressing insulin with a TZD on LH pulse characteristics and indexes of insulin secretion/sensitivity will also be assessed.
