This subproject is one of many research subprojects utilizing the
resources provided by a Center grant funded by NIH/NCRR. The subproject and
investigator (PI) may have received primary funding from another NIH source,
and thus could be represented in other CRISP entries. The institution listed is
for the Center, which is not necessarily the institution for the investigator.
We are developing a moment analysis technique which can be used to determine protein number and clustering in solution as well as in live cells. The theory has been developed for both photon counting and analog systems. Photon counting systems are able to recover absolute number and brightness information with high accuracy, whereas analog systems can potentially recover accurate values provided the detector can be properly characterized and remains stable during the course of the measurement. Lastly, immobile and mobile fractions can be distinguished as well.
