This subproject is one of many research subprojects utilizing the
resources provided by a Center grant funded by NIH/NCRR. The subproject and
investigator (PI) may have received primary funding from another NIH source,
and thus could be represented in other CRISP entries. The institution listed is
for the Center, which is not necessarily the institution for the investigator.
This core will provide proteomics analytical services and technical assistance to the COBRE investigators. The core will be directed by Dwight Matthews, Departments of Medicine & Chemistry and by Gary Ward, Department of Microbiology & Molecular Genetics. Dr. Matthews is responsible overall for the core and will focus on the mass spectrometry aspects of the core. Dr. Matthews has a several-decade long
history in mass spectrometry and directs mass spectrometry instrumentation in several locations at UVM. Dr. Ward will focus on protein isolation and separation techniques. Clearly, the power of mass spectrometry is lost if the samples to be measured cannot be isolated in a viable form. In addition, Dr. Ward will be
responsible for 2-dimensional (2D) gel electrophoresis separation of proteins. This technique is valuable as a stand-alone technique in the core. Dr Ward has extensive experience in protein purification and separation, including a variety of chromatographic techniques and 2D gel electrophoresis. Technical expertise will also
be available specifically through this core in the form of a technical person who will be responsible for both running samples by mass spectrometry and for preparing samples and 2D gel separations. The technical person will handle many of the hands-on tasks that the investigators will not have the time and expertise to
manage. The technical person will also assist in training interested investigators and students how to perform the techniques required to run their own samples. Procedures will be in place for investigators to assure that they will obtain results that are optimal for their experiments and protocols. Both Drs. Matthews
and Ward will work directly with the investigators at all stages from planning experiments to interpreting results. The proteomics techniques are complicated and fraught with subtle complexities, and the core will assist users in this regard.
