This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The actin cytoskeleton functions in a variety of important cellular processes including endocytosis, cell motility, cell morphology and organelle transport. The actin cytoskeleton is a dynamic network of filaments and associated proteins that undergoes disassembly and reassembly in response to various cellular and extracellular signals. The Ser/ Thr kinases, Ark1 and Prk1, were recently found to couple the dynamic processes of the actin cytoskeleton to endocytosis in Saccharomyces cerevisiae. The mammalian homologs of these kinases, AAK1 and GAK, are also implicated in the control of endocytosis in mammalian cells. To elucidate the regulatory mechanisms of Ark1/ Prk1 in the actin cytoskeleton assembly and endocytosis, we are exploiting chemical-genetic/ proteomic strategies to identify kinase targets of Ark1 and Prk1.
