This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The technology of microarray analysis of gene expression is not yet being optimally applied to nonhuman primate research. One widely available tool for microarray analysis are the Affymetrix gene chips which can analyze over 15,000 mRNAs per chip. However, because the Affymetrix chips are based on short oligonucleotides for which a single base mismatch will prevent hybridization, it is generally thought that Affymetrix chips are not suitable for cross-species hybridization, even for species as close as monkeys and humans. In this project, which is a collaboration between the ONPRC and the University of Nebraska, construction of specific rhesus monkey microarray chips will be facilitated by generating rhesus cDNA sequences using targeted PCR techniques. To date, over 3,000 sequences have been generated and deposited to genbank. The next step will be to combine these sequences with other available rhesus DNA sequences to design rhesus oligonucleotide chips that will be commercially available to the entire primate community.
