Funds to purchase a stereomicroscope and a preparative centrifuge are requested. The preparative centrifuge will be used to harvest cell cultures containing recombinant proteins and to separate recombinant proteins from cellular debris. The stereomicroscope will be used to observe protein crystals during crystallization setups. The abilities to harvest cells, separate recombinant proteins and most importantly to observe protein crystals is fundamental for the success of the NSERC Discovery research project. The focus of the research program is to use macromolecular crystallography to characterize deubiquitination in yeast including the determination of tertiary structures of ubiquitin specific proteases and the characterization of the mechanism of interaction between ubiquitin specific proteases and their target proteins. This equipment will be made available to other members of the department and will be instrumental in the training of highly qualified personnel.
