B-cell Memory is crucial for rapid, effective and highly targeted immune responses. Long lasting immunity, be it following natural infection or through vaccination, is dependent on the optimal development and location of memory B-lymphocytes. Three subsets of mature B-cells contribute to B-cell memory responses; CD27+ Memory B-cells, plasmablasts, and plasma cells. Through transcriptional profiling, we have identified an important novel surface molecule, Semaphorin 4C, that is specifically expressed in the development of B-memory lineage cells. Our preliminary evidence demonstrates that Sema 4C may be important in B-cell development and in the interaction of circulating B-mem subsets within the matrix of peripheral tissues. Our research program aims to understand the developmental influences on B-cells in airway mucosa. In this proposal, using our well defined murine models, we will build the foundation to elaborate the functional role of Sema 4C in the development of B-cell memory responses.
