Intensive work is now being performed both in the academic and commercial sectors to develop methods of using light scattering in body tissues for non-invasive diagnosis of specific conditions such as urinary tract infections. These methods are highly dependent on an accurate measurement of tissue oxygenation. A major challenge for oxygenation measurements that are based on infrared absorption spectroscopy is to properly account for the effect of various tissue layers in the body on such measurements. This proposal is aimed at researching and developing a precise mathematical model that corrects IR based oxygenation measurements on various tissue layers between the skin and the muscle being measured. Our approach will be to first experimentally identify the primary factors that affect oxygenation measurements from standard infrared (IR) absorption spectroscopy and therefore to investigate the role of light propagation through different tissue layers on these spectroscopic measurements. Based on these investigations, we will develop a mathematical model that accounts for the effect of light propagation in each layer. The goal is to provide a reliable way to calibrate IR spectroscopic measurements based on the geometry of different tissue layers in patients.
