Canid herpesvirus 1 (CHV-1) is an important veterinary pathogen that causes respiratory and genital infections in dogs. In puppies, it can lead to a generalized infection and death. In adult dogs, the virus is associated with ocular and respiratory disease. It has also been associated with infertility and abortions. There are limited treatment options. CHV-1 is a member of the genus Varicellovirus. It only infects dogs and dog-like animals such as wolves, coyotes, foxes and jackals. Despite the importance of this pathogen in veterinary medicine, very little is known about the biology of CHV-1. The objective of this research program is to understand the molecular and cellular biology of CHV-1 infection. We will target two critical aspects of CHV-1 infection: 1) How the virus enters and traffics within highly specialized polarized cells such as those found in the respiratory track. These cells have two sides with very different functions, and mechanisms in place that govern which molecules ultimately go to which side. 2) We will also study what controls how viral genes are turned on and off. The results generated will be of particular interest to those who have undertaken to use CHV-1 as a vector for dog-specific therapeutic approaches. The information generated should reveal critical aspects of CHV-1 infection, an understudied veterinary pathogen. It will also likely generate new fundamental knowledge regarding the infection of polarized epithelial cells and strategies of viral gene expression.*****
