The proposed research program will challenge the existing principles of chemistry in order to find new therapeutics. The Yudin lab will demonstrate that taking macrocycles away from their equilibrium states is an innovative new strategy for creating bioactive structures. The emerging understanding of complex macrocycles will enable the construction of new classes of bioactive molecules. The Yudin lab will control the specificity of biomolecular interactions by targeting weakly nucleophilic residues in protein binding sites using novel boron-containing compounds. This knowledge will lead to a radically different approach to the discovery of new bioactive molecules and their subsequent translational studies.
