The objective of this project is to seek for the method to induce new bone in vivo which is adequately adapted to the shape of bone defect area by using scaffolds which has capability to form the same shape as mentioned as above with BMP and/or host origin cells, then implanted into the living tissue. The project has succeeded to make the EMA resin-BMP composites which induce new bone, three dimensional detection of the shape which was formed by resin material and rapid screening system for the cytotoxicity of materials by using the marker gene of GST. On the surface of the newly innovated resin that is containing PLGA, BMP was combined. This system revealed the stable new bone formation, and carrier such as PLGA showed excellent result as carrier of BMP, and it was also proven to be able to substitute the dextrin for the polylactic acid. Moreover, three dimensions of the bone induction and changes the passing time were clarified for the first time in the world wide by a high-speed, X- … More ray CT scan and also showed the 3-D image of new bone formation by thin resin film with BMP which is the main object of this experiment. Establishment of the bioassay system of self-luminescence using luciferase gene co-transfected with GST into the cultured cells to detect the cytotoxyty is remarkable achievement. However, to cultivate cells on the EMA resin could not have good result. Same kind of cultivation was performed by the organ culture system of undifferentiated skeletal muscle, which has been commonly experimented in this group. Eventually, this system also showed poor aggregation of tissues. Nonetheless, the information of these experiments showing was extremely important for the future work and the possible potential molecules containing in the EMA resin system may cause the inhibition of the mesenchymal tissue proliferation. Systems obtained through this project, such as 3 dimensional scaffolds with BMP, will attribute to developing new tissue engineering methods, and this project will be continuously experimented from now on.
