Camalexin (3-Thiazol-2'-yl-Indol) wird in Arabidopsis als Antwort auf Pathogenbefall gebildet und verleiht der Pflanze erhöhte Resistenz gegenüber beispielsweise Alternaria brassicicola. In diesem Projekt soll die bisher nur unzureichend verstandene Camalexin-Biosynthese untersucht werden. Zunächst soll durch retrobiosynthetische NMR-Analyse bzw. spezifische Markierungsexperimente bestimmt werden, ob Indol-3-glycerinphosphat, Indol oder Tryptophan den Ausgangspunkt vom Grundstoffwechsel darstellt. Sollte die Biosynthese Tryptophan-abhängig verlaufen, so kann die mögliche Vernetzung mit der Indol-Glucosinolat- und Auxin-Biosynthese untersucht werden. Die Funktion des wahrscheinlich an der Camalexin-Biosynthese beteiligten Cytochrom-P450 Enzyms CYP71B15 (PAD3) soll durch heterologe Expression und die Analyse von pad3-Mutanten geklärt werden. Von der Identifizierung biosynthetischer Zwischenstufen ausgehend sollen weitere biosynthetische Gene identifiziert und charakterisiert und deren Regulation untersucht werden. Die Aufklärung der Camalexin-Biosynthese in Arabidopsis wird die Grundlage für ein besseres Verständnis der induzierten Pathogenabwehr der Pflanzen bilden.
