Identification of the major gene for resistance to Soybean dwarf virus

利用病毒诱导基因沉默（VIGS）抑制了大豆矮缩病毒（SbDV）主要抗性基因Rsdv1的表达。建立了用酶联免疫吸附法检测SbDV耐药性而不采用症状观察的方法。利用该方法，可以对由于VIGS引起的症状而无法检测到SbDV症状的基因沉默大豆品系进行抗性评估。候选Rsdv1基因的VIGS未显著改变sbdv侵染植株的百分比，说明VIGS不适合鉴定Rsdv1，或者需要额外的邻近基因才能产生抗性。

The expression of the candidate Rsdv1 gene, the major Soybean dwarf virus (SbDV) resistance gene derived from the soybean cultivar 'Wilis', was suppressed by virus-induced gene silencing (VIGS). An evaluation method for SbDV resistance was developed, in which SbDV was detected by ELISA instead of symptom observation. This method enabled resistance evaluation of gene-silenced soybean lines whose symptoms of SbDV are not detectable due to the symptoms caused by VIGS. VIGS of the candidate Rsdv1 gene did not significantly change the percentage of SbDV-infected plants, suggesting that VIGS was not suitable for identification of Rsdv1, or that additional neighboring genes were necessary for resistance.
