Tocopherol (Vitamin E) stellt einen wichtigen Ergänzungsstoff für die menschliche Ernährung dar. Es kann nicht von Tieren synthetisiert werden, sondern muss in der Nahrung aus Pflanzen aufgenommen werden. In Pflanzen wird die Synthese von Tocopherol durch oxidativen Stress induziert, und man nimmt an, dass es am Abbau reaktiver Sauerstoff-Spezies beteiligt ist. Die Identifizierung von zwei Tocopherol-defizienten Arabidopsis Pflanzen (vte1, vte4) zeigte jedoch, dass der Verlust von Tocopherol nur zu einer geringen Beeinträchtigung des Wachstums führt. Diese Mutanten und eine dritte Mutante (hpt1), die wahrscheinlich einen Defekt in der Homogentisat-Phytyltransferase trägt, sollen im Rahmen dieses Projekts zur Aufklärung der Rolle von Tocopherol beitragen. Um die Frage zu klären, ob der Verlust von Tocopherol in vte1 durch andere Antioxidantien (z.B Vitamin C/Ascorbat, Glutathion, Xanthophylle) kompensiert wird, werden Doppelmutanten von vte1 mit vtc1 (Ascorbat-defizient), cad2 (Glutathion-defizient) und npq1 (Zeaxanthin-defizient) erzeugt und biochemisch analysiert. Wir werden die Rolle von Tocopherol bei der Degradation von Singulett-Sauerstoff aus Photosystem II untersuchen. Weiterhin werden wir mit vte1 Expression Profiling mit Oligonukleotid-Chips durchführen, um Hinweise auf die Existenz neuer Tocopherol-abhängiger Signaltransduktionswege oder Stoffwechselwege oder Stoffwechselwege zu erhalten.
