In order to confirm that isoprenylation of tryptophan residues is a universal posttranslational modification, I developed a cell-free assay system by which the tryptophan residue in the ComX pheromone precursor can be isoprenylated by themodifying enzyme, ComQ, with isoprenyl pyrophosphate and magnesium ion.
