神経膠腫症例の摘出サンプルより抽出したRNAを用いたノザンブロッティングによる解析で、最も未分化な膠芽腫症例の一部でACS 2 mRNAの発現が消失していることを発見し、分化度の高い星細胞腫や退形成性星細胞腫では発現の消失が認められないことが明らかとした。また星細胞系の腫瘍以外の稀突起膠種、上衣腫、神経節膠腫などではその発現は認められなかった。神経膠腫樹立細胞株ではACS2の発現は認められなかった。ACS2(LACS5)には2つのルシフェラーゼ類似領域があり、この内の一つ目のルシフェラーゼ類似領域内に合成ペプチドを設定し、これを合計3匹のウサギに投与して、抗ACS2抗体を作成した。3匹分の血清抗体は、western blottingにより、いずれの血清抗体もACS2を認識することを確認した。この抗体を用いて免疫染色を行った。正常脳組織では神経細胞と膠細胞ともに染色された。astrocytoma grade 2及びgrade 3の症例では、northern blotにて発現が確認されたが、免疫染色にてgemistocyteを含む腫瘍細胞の細胞質が染色された。この結果はACS2がミトコンドリア、ペルオキシゾーム、小胞体に発現していることに合致した結果であった。astrocytoma grade 4の症例では、Northern blotで陰性のものは腫瘍細胞は染色されず、陽性のものは腫瘍細胞が染色された。陰性の症例は壊死組織が顕著である傾向が認められた。
