RESRCH ON THE CONSERVATION OF AVIAN WAS CONDUCTED.AT PRESENT, AVIAN GENETIC RESOURCES ARE CONSERVED AS FLOCK SINCE IT IS NOT POSSILBLE TO CONSERVE FERTILIZED EMBRYOS FOR MORE THAN 2-3 WEEKS WITHOUT IMPAIRING HATCHABILITY.NEW STRATEGY TO CONSERVE AVIAN GENETIC RESOURCES NEEDS TO BE DEVELOPED.TO ACCHIEVE THE GOAL, GERM-LLINE CHICMERAS WERE PRODUCED BY TRANSFERRING PRIMORDIAL GERM CELLS (PGC).REDULTS OBTAINED IN THE PRESENT RESEARCH PROJECT WAS AS FOLLOWS : 1) NUMBER OF THE CIRCULATING PGC WAS HIGHEST AT STAGE 14 AND DECREASED THEREAFTER.2) TOTAL NUMBER OF CIRCULATINF PGC WAS ESTIMATED TO BE 807 AND 857 AT STAGE 14 AND 15, RESPECTIVELY.3) THE REASON FOR THE CONSIDERABLE FULCTUATION OF THE PCG CONCENTRATION IN EARCLY CHICK EMBRYO WAS PRIMARIRILY ATTRIBUTED TO THE FEMALE THAT PRODUCE EGG.4) GERM-LINE CHIMERAS WERE PRIDUCED BY TRANSFEERING CRYOPRESERVED GONADAL PGC (GPGC) COLLECTED FROM 5-DAY-OLS CHICK EMBRYO.5) NEW METHOD OF COLLECTING COCULATING PGC USING FILTRATION METHOD WAS DEVELOPED.
对鸟类遗传资源的保存进行了研究。目前,由于受精胚胎保存时间不可能超过2-3周而不影响孵化率,因此,鸟类遗传资源的保存基本上是以群的形式进行的。需要制定新的鸟类遗传资源保存策略。为了实现这一目标,用原生殖细胞(PGC)转化法生产鸡胚,获得的红色素如下:1)循环PGC的数量在第14阶段最高,此后下降。2)在第14和15阶段,循环PGC的总数估计为807和857,3)早期鸡胚PCG浓度的合理波动主要归因于产蛋的雌性。4)生殖-从5日龄鸡胚中提取冷冻保存的生殖腺PGC(GPGC),经冻存后进行细胞移植,制备线嵌合体。5)建立了用过滤法提取融合PGC的新方法。
