1.Molecular structure of membrane lipid peroxidesUsing LC/APCI-MS, 7-ketocholesterol and 7-hydroxycholesterol were identified in skeletal muscle and kidney of rats. Phosphatidyocholine (PC) monohydroperoxide (PC 16:0/18:2-OOH and PC 18:0/18:2-OOH) were identified in rat heart and human plasma by LC/ESI-MS.2.Quantitative analyses of oxysterol by HPLC-UV and PCOOH by HPLC-CL were developed.3.Accumulation of oxysterols in lung of victims with paraquat poisoning was associated with pulmonary fibrosis. Consecutive administration of paraquat to rats induced enhanced cholesterol peroxidation and lung injury.4.Chronic ethanol feeding induced accumulation of oxysterols in skeletal muscle and liver. Plasma PCOOH is elevated in alcoholic patients with liver disease suggest the involvement of lipid peroxidation in alcohol-induced liver damage.Thus, we have shown not only oxysterol but PCOOH are prominent markers of oxidative stress.
1.用LC/APCI-MS技术,在大鼠骨骼肌和肾脏中鉴定出7-酮胆固醇和7-羟基胆固醇。采用LC/ESI-MS法测定了大鼠心脏和人血浆中的磷脂酰胆碱(PC)单氢过氧化物(PC 16:0/18:2-OOH和PC 18:0/18:2-OOH)。百草枯连续染毒可引起大鼠胆固醇过氧化反应增强和肺损伤。4.慢性乙醇喂养可引起大鼠骨骼肌和肝脏中氧化固醇的积累。酒精性肝病患者血浆PCOH升高提示脂质过氧化参与了酒精性肝损伤,因此,我们发现不仅氧固醇而且PCOH都是氧化应激的重要标志物。
