Effects of Growth Factors and Carcinogens on the Development of Odontogenic Tumors

本研究采用裸鼠皮下移植ddY鼠牙胚的方法，观察了生长因子TGF-α和致癌物Trp-P-2对ddY鼠牙胚发育的影响，并采用免疫组化方法检测了5组小鼠的PCNA表达。A组（对照组）、B组（DMSO组）、C组（TGF-α组）、D组（Trp-P-2组）和E组（TGF-α+Trp-P-2组）。从历史上看，在C、D和E组中，观察到牙源性上皮细胞显著增殖。D、E组牙源性上皮细胞角化，C组未见角化。此外，在E组中，在角化牙源性上皮细胞区域中观察到囊肿样间隙。免疫组化结果显示，C、D、E组牙源性上皮细胞的阳性表达率明显高于A、B组。本研究结果提示，生长因子TGF-α和致癌物Trp-P-2可促进小鼠牙胚移植后牙源性上皮细胞的增殖和角化。

In the present study, the effects of both growth factor TGF-α and carcinogen Trp-P-2 on the development of organ-cultured ddY mouse tooth-germ transplanted into the subcutaneous tissue of nude mice were investigated historically and immnohistochemically (PCNA) in the following 5 Groups of mice : Group A (Control), Group B (DMSO), Group C (TGF-α), Group D (Trp-P-2) and Group E (TGF-α+Trp-P-2). Historically, in Groups C,D and E, a marked proliferation of odontogenic epithelial cells was observed. In Groups D and E, keratinization of the odontogenic epithelial cells was found, but it was not observed in Group C. Moreover, in Group E, a cyst-like space was observed in the area of keratinized odontogenic epithelial cells. Immunohistochemically (PCNA), in Groups C,D and E, the positivities of the odontogenic epithelial cells were significantly higher than in Groups A and B. The results of the present study suggest that proliferation and keratinization of odontogenic epitherial cells of transplanted organ-cultured tooth-germs in mice are promoted by growth factor TGF-α and carcinogen Trp-P-2.
