Development of BrdU releasing polymer and its use on radiosensitization of brain tumors : experimental study.

乙烯-醋酸乙烯酯共聚物是一种方便的药物缓释载体。BrdU是胸腺嘧啶核苷类似物，可掺入S期细胞，增强细胞对辐射和紫外线的敏感性。我们开发了能够持续释放BrdU的BrdU聚合物。我们将该聚合物应用于大鼠9 L胶质瘤的皮下移植，并检测了BrdU聚合物的放射增敏作用。然而，我们没有发现放射加聚合物治疗的大鼠和仅用放射治疗的大鼠之间的肿瘤大小的差异。BrdU聚合物在体内胶质瘤中没有放射增敏作用的原因正在调查中。

Ethylene vinyl acetate copolymer is a convenient vehicle capable of sustained drug release. BrdU,thymidine analogue, is incorporated into S-phase cells and enhances susceptibility of cells to irradiation and UV.However, BrdU was not used generally because of its systemic toxicity. We developed BrdU polymer capable of sustained BrdU release. We applied this polymer to subcutaveous rat 9L glioma and examind radiosensitizing effect of BrdU polymer. However, we did not find difference in tumor size between rats reated by irradiation plus polymer and those treated only by irradiation. The reason why BrdU polymer did not show radiosensitizing effect glioma in vivo is under investigation.
