项目摘要

The concept of neuronal regeneration has been rapidly changed from the past regenerative medicine. Recently neuronal differentiation and transplantation could be possible using by embryonal stem cell. Regeneration is much interested in the clinical field, so we studied for the possibility of neuronal regeneration in ischemic bran using bone marrow transplantation.Experiment of ischemic brainWe succeeded in modeling complete occulusion of ipsilatenal middle cerebral artery.Differentiation of bone marrow stem cells (BMC)BMS including the hematopoietic and mesenchymal cells could be differentiated into neuronal direction by adding PDGF-BB and/or b-FGF.Sorting and neuronal expression of CD34CD34- and CD34+ cells of lineage depleted BMC were sorted and neuronal differentiation showing lin-CD34-adult murine BMC were found by immunocytochemistry and PCR.Improvement of ischemic brainThe ischemic region was improved by transplantation of BMC.
