Human artificial chromosome (HAC) is maintained stably with one copy in the cell. In CENP-B gene deficient mouse cell, HAC exists with multi copy and becomes unstable. When HAC with CENP-B gene is transferred into CENP-B gene deficient cells, the stability of HAC was recovered. To clarify mechanism for stability of HAC in CENP-B deficient cell, HAC with unknown gene was introduced. I demonstrate that several nucleoproteins are concerned to centromere/kinetochore function with CENP-B.
