Examination of an adjuvant effect to the cancer cell of CpG in vivo

(2010)(1)B16黑色素瘤购自日本理健公司，培养扩增。(2)给WT小鼠(C57BL/6)注射抗癌药物5-FU，7天后进行流式细胞术和HE染色，观察骨髓抑制情况。(3)给予CpG佐剂，用ELISA法观察炎性细胞因子的产生。(2011)(1)B16黑色素瘤皮下接种，证实移植模型。外周血中髓系上皮细胞(MDSC)占8%，注射5-FU后7d，MDSC减少4%。联合应用5-FU和CpG后，MDSC增加20%。

(2010)(1) B16 melanoma were purchased from RIKEN, incubated for proliferation. (2) Anticancer drug, 5-FU were administered to WT mice(C57BL/6), and myelosuppression was observed, so called nadir, at 7days Flowcytometry and HE staining. (3) An adjuvant CpG were administered, the production of inflammatory cytokine were observed by ELISA. (2011) (1) B16 melanoma were inoculated s.c., as a result, confirmed engrafted model. Myeloid derived suplessive cells(MDSC) resident in peripheral blood at 8%, but 7days after 5-FU injection, MDSC were decreased at 4%. On the other, 5-FU and CpG combined injected, MDSC were increased at 20%.
