We have finished the construction of HAC701 fusion protein for rice protoplast and callus, but was never substantially tested. This will be continued as budget becomes available for this project. We have produced the hac701/wrky45 double mutant and will be ready for genotyping with the help from Plant Epigenetics Unit (OIST). This double mutant will clarify some of the reviewer’s questions on the specificity of CRISPR/Cas9 targeting used to silence HAC701. In summary, we have provided evidence for the genetic interaction of HAC701 and WRKY45. Also, we have confirmed that H3K9ac is possibly catalyzed by HAC701 acetyltransferase. Lastly, we found enrichment of WRKY45 sequences in H3K9ac fraction adding more evidence of the possible physical interaction of HAC701 with WRKY45 locus. The manuscript has been accepted for publication at Communications Biology provided major revisions be supplied.
