The proposed project aims to investigate the contribution of a complex signaling network comprising Calcineurin B-like proteins (CBLs) and their interacting protein kinases (CIPKs) to calcium signaling processes in plants. In general, this project is focused on protein phosphorylation processes as important steps in generating and decoding calcium signals. Specifically, this project aims to: (i) investigate protein phosphorylation as functional principle regulating CBL/CIPK complex activity, (ii) investigate the role of CBL/CIPKs in regulating ABA-responsive transcription factors, (iii) investigate the interconnection of CBL/CIPKs with PP2Cs in regulating ABAperception, (iv) investigate the interplay of CBL/CIPKs with CDPKs and SnRK2s in regulating ion transport, (v) identify and characterize CBL/CIPK targets by the nonbiased identification of phosphorylation substrates by comparative phosphoproteomics analyses of quintuple cbl mutants, and (vi) analyze the dynamics of Ca2+ concentration in response to ABA or cold at high tissue-specific and sub-cellular resolution.
