VISUALIZING ASSOCIATION OF N-RAS IN LIPID MICRODOMAINS

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Two photon fluorescence microscopy on giant unilamellar vesicles is used to follow the insertion of a fluorescently(Bodipy)labelled and completely lipidated (bi-hexadecylated) N-Ras protein into heterogeneous lipid bilayer systems. The bilayer consists of the canonical raft mixture POPC, sphingomyelin and cholestero, which -depending on the concentration of the constituents-separates into liquid-disordered(ld), liquid-ordered(lo) and solid-ordered(So). The result of this experiment will provide the direct evidence of the preferable partitioning of N-Ras into specific domains.
