This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The Asf1/Hira complex are involved in chromatin assembly and also contribute to the formation of heterochromatin and telomeric silencing and thus contribute to cell cycle exit in senesent cells. This process is abbarantly regulated in cancer. We have prapred crystals of the Asf1/Hira complex that should be suitable for X-ray structure determination using the nascent Asf1 as a search model in molecular replacement. These native crystals show diffraction that can easily be processed and scaled but the diffraction extends to relatively low resolution using a home X-ray sources under cryogenic conditions. Attempts to work out a molecular replacement using a low resolution data set (4 A)has been unsuccessfully thus far. Therefore, collecting a higher resolution data set for each of these crystals is highly desirable. Given the competitive nature of this project, rapid access to CHESS is necessary.
