NMR STUDY OF THE TITRATION OF YGGX WITH GALLIUM III

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. YggX is an 11 kDa protein found in Salmonella enterica as well as in several other diverse bacteria that has been implicated in iron trafficking and in protecting the cell from oxidative damage. Circular dichroism studies on purified YggX have shown that this protein undergoes a conformational change upon exposure to iron III as well as to gallium III. In order to understand this conformational change in more detailed structural data is needed. An N15 labeled sample of YggX protein will be titrated with a sample of gallium III in order to obtain detailed structural data. The results of this experiment will then be used in order to decide what forms of YggX should be used for determining a solution structure
