This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Yeast have one protein having significant structural and functional similarity to higher cell histone H1. We hypothesize that, as for the core histones, the "tail" and (for yeast H1 only) internal "linker" peptides represent protein-protein interaction modules whose activity will prove to be controlled by specific posttranslational modifications. We seek to identify such interacting partners. The genome-wide affinity-tagging project has identified several candidates, but their validity is difficult to assess at present. Thus, we propose several complementary approaches, including a genome-wide 2-hybrid experiment.
