Program Director/Principal Investigator (Freed, Jack, H.): Specific Aims The overall mission of ACERT (the National Biomedical Resource for Advanced Electron Spin Resonance Spectroscopy) is twofold: (1) to take advantage of the latest developments in ESR techniques to provide state-of-the-art facilities that enable molecular biologists to perform studies that advance biomedical research, and (2) to provide advanced ESR-based services to scientists working at the frontiers of molecular biology (taken in its most general sense), with long-term benefits to the biomedical research community as a whole. A special opportunity for the ACERT program is provided by upgrades to the ESR spectrometers, currently being completed. ACERT will promote the application of cutting-edge instrumentation and techniques to some of the most challenging questions confronting molecular biologists. We will use the expertise of ACERT personnel, the administrative leadership team, and the external scientific advisory group to provide support to molecular biologists using the facility. Our primary aim in this supplementary request is to maximize spectrometer live time available for the ACERT Resource users to enable accepting and processing more requests and to expand the range of amenable systems by (1) implementing round the clock cryogen-free operation of the spectrometer and (2) to further enhance its performance by installing commercial high-sensitivity sample resonators.
