Novel intracellular signaling mechanisms: RACK1 interacts with GRK2 and arrestin

Our laboratory is focused on studying the mechanisms regulating the thromboxane A2 (TXA2) receptor, of the family of G protein-coupled receptors (GPCRs). These receptors are protein present at the surface of all existing cell types. They constitute the largest single class of receptors. They transmit signals from neurotransmitters, odorants,food (taste), light, and hormones from the extracellular environment to the inside of the cells. Signal transduction of GPCRs is highly regulated intracellularly. We are studying these regulatory mechanisms in our laboratory using the TXA2 receptor as a model. We have recently identified novel interactions between a protein called RACK1 and two other proteins referred to as GRK2 and arrestin. GRK2 and arrestin are known to initially decreased the receptor response. However, arrestin can also promote various intracellular signals. Our NSERC research program will try to elucidate how the RACK1/GRK2 and RACK1/arrestin interactions are involved in the regulation of this very important family of receptors.
