Development of BrdU releasing polymer and its use on radiosensitization of brain tumors : experimental study.

乙烯醋酸乙烯共聚物是一种方便的缓释药物载体。胸苷类似物BrdU被纳入s期细胞，增强细胞对辐射和紫外线的敏感性。然而，BrdU由于其全身毒性而未被广泛使用。我们开发了能够持续释放BrdU的BrdU聚合物。我们将该聚合物应用于大鼠皮下9L胶质瘤，观察BrdU聚合物的放射增敏作用。然而，我们没有发现放射加聚合物治疗的大鼠与仅放射治疗的大鼠之间肿瘤大小的差异。BrdU聚合物在胶质瘤体内没有放射增敏作用的原因还在研究中。

Ethylene vinyl acetate copolymer is a convenient vehicle capable of sustained drug release. BrdU,thymidine analogue, is incorporated into S-phase cells and enhances susceptibility of cells to irradiation and UV.However, BrdU was not used generally because of its systemic toxicity. We developed BrdU polymer capable of sustained BrdU release. We applied this polymer to subcutaveous rat 9L glioma and examind radiosensitizing effect of BrdU polymer. However, we did not find difference in tumor size between rats reated by irradiation plus polymer and those treated only by irradiation. The reason why BrdU polymer did not show radiosensitizing effect glioma in vivo is under investigation.
