X-RAY STUDIED OF REL-MTB

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. We have crystallized Relmtb, a Rel/SPoT homolog from M tuberculosis that modulates the stringent response in that bacterium. Knockouts of this gene demonstrate increased susceptibility to antibiotics. We have collected native data to 2.3 Ang resolution. However, due to the very small (<50 um) crystal size, overall I/sigma(I) for these data are low (~5) and Rsym is rather high (0.16). We expect that synchrotron data will ameliorate these problems, and will also allow us to solve the structure by MAD or MIRAS.
